BLAST Activity #1: Use of a database to determine differences in the base sequence of a gene in two species.
1. Go to ncbi.nlm.nih.gov and search all databases for HBB. HBB is the abbreviation for beta hemoglobin. 2. Scroll down on the left side and select "Nucleotide" 3. On the right, select "Homo sapiens". 4. Scroll down (result #76) to find the search result that is 1605 bp long (bp = base pairs) and select this one. 5. Select "Run BLAST" on the right side. 6. In the "Organism" box, type "Pan troglodytes"; this is the scientific name for a chimpanzee. 7. At the bottom, hit the BLAST button...and wait. 8. Look at the results table and see what percent of similarity is reported between the two sequences. 9. Pick one other animal to compare to the human HBB sequence. |
BLAST Activity #2: Use of a database to discover gene mutation conditions
1. Go to a nucleotide BLAST (blastn). 2. Be sure your organism is set to Homo sapiens. 3. Select your assigned gene sequence below and copy it into the large sequence box at the top of the page. 4. Hit the BLAST button at the bottom. 5. Select the "Accession" link in the top match to see more about this gene. You can then click on the "More about the ________ gene" link on the right side to learn more about what conditions are associated with this gene. 6. Do this for your assigned sequences below. For each, tell (simply citing all sources): a) The name of the disease caused by the variant. b) The chromosome on which it is located. c) The specific mutation. d) The protein affected and the direct effects. e) Symptoms of the disease. f) Treatment of the disease. g) Current research on the disease.
